Synovate, a leading global market intelligence company, today released information from the 2005 Synovate PAX media survey that showed rises in wealth, product and service ownership and improvements in lifestyle across Singapores affluent residents. The Synovate PAX survey tracks media, prosperity and influence in eleven markets across the Asia Pacific region – Singapore, Hong Kong, Korea, Malaysia, Thailand, Indonesia, Philippines, Taiwan, India, Australia and Japan. Wanda Gill, Media Director for Synovate in Singapore said that the affluent of Singapore were enjoying high confidence and spending power. In just the last few months, our constantly-updated tracking has enabled us to watch Singapores ever–increasing confidence translate into higher product ownership, increasing purchase intentions and improving lifestyle. The Synovate PAX results for 2005 show that the average monthly household income of the Lion Citys affluent residents has risen from US$4,826 to US$5,456 – a 13% relative change. With greater household wealth, these Singaporeans are rewarding themselves with purchases of top tier luxury and consumer items and steady increases in property ownership and investment, Ms Gill said. The Synovate PAX survey allows marketers to get a full picture of Singapores affluent consumers – what they watch, read, buy and how they feel about advertising and branding – enabling them to target their marketing efforts. Ms Gill said that the 2005 results painted a picture of Singapore as the quiet powerhouse in relation to other markets in the region. While Hong Kongs wealthy are conspicuous consumers and buy items for their fashion status, Singaporeans tend to be more considered in distributing the fruits of their labours. Singapores elites still spend – and spend a lot – but our data shows they wont get their wallets out on a whim. Marketers must appeal to this lack of pretension and realise Singaporeans are not as status-conscious as people from Hong Kong or as acquisitive as the wealthy of other South East Asian economies which are less mature. Singaporeans are comfortable in their skins and are more likely to buy to make their lives easier, Ms Gill said. The Synovate PAX survey delves into peoples attitudes to brands and advertising via a series of psychographic statements. Some of the findings for Singapore were: * 11. 5% of Singapores elites agreed with the statement that they are among the first to buy a new product or gadget. This is below the regional average, showing that Singaporeans are less in a rush than other markets, waiting to see if the item fits into their lifestyle. More than two thirds of Singaporeans agree that paying for quality is worthwhile. * 46. 9% wealthy Singaporeans prefer to buy well known brands, well over the regional average of 41. 1%. * Singaporeans show a focus on lifestyle with 41. 5% agreeing that a home with modern appliances is important to them – this compares to the regional average of 38. 7% and 28. 9% in Hong Kong. Theres an indulgent streak across affluent Singaporeans, with one of the highest levels of agreement to the statement I sometimes like to treat myself to something special. 8. 2% of PAX respondents agreed in Singapore, with only Sydney-siders more indulgent (62%). The regional average is 48. 5%. Synovate PAX also covers product and service ownership, purchase intention over the twelve months to June 2006 and lifestyle. * As a major regional business hub of Asia, it would be expected that a great deal of business travel originates here. However, from June 2004 to June 2005, we have seen a 22% increase in Singapores affluent taking off on one or more business trips. Around 28% of these PAX respondents travel for business. But Singapores elite still find time to relax – more than 50% have taken one or more leisure trips in the year to June 2005 too. * An all-time high number of 38. 6% of affluent Singaporeans now own laptops or notebooks, up from 31. 9% in June 2004. * Likewise, ownership of a mobile phone with Internet access hits an all-time high – it stands at 37. 7% and shows steady increases for every quarter measured. This continuing trend is good news for mobile manufacturers and 3G networks and service offering planners. Indeed, more than 92% of affluent Singaporeans own a mobile with or without Internet access – they are here to stay and all these people will upgrade as mobile content becomes more and more compelling. * Digital dominance seems more and more inevitable as we observe relentless increases in Singaporean ownership of devices like MP3 players (up to 23. 1%, a rise of a quarter over the year before), digital video cameras (43. 7% now own these, up from 35. 9%) and digital still cameras (more than half of affluent Singaporeans own these an increase from 41% in 2004). Indeed, the love ffair with digital is truly blossoming. * Its fair to say that the marketers of LCD or plasma televisions have enjoyed a good year. They close to doubled their market size, with the number of wealthy Singaporeans already owning one of these TVs rising from 7. 6% to 12. 7%. And theres still a great deal of upside, with 10% expressing the intention to purchase this top tier item in the next 12 months. * Car ownership impressively jumped for the second consecutive year. Tragic Hero in The Crucible What is a tragic hero? The most well known definition of a tragic hero comes from the great philosopher, Aristotle. When depicting a tragic hero, Aristotle states "The change in the hero's fortunes be not from misery to happiness, but on the contrary, from happiness to misery, and the cause of it must not lie in any wickedness but in some great error on his part.† In addition, he explains the four essential qualities that a tragic hero should possess, which are goodness, appropriateness, lifelike, and consistency. All of these necessities help to classify the character of John Proctor as the tragic hero of the play. Not only does he unfortunately fall to desolation, but also his character captures the sympathy and pity from the audience or readers. A tragic hero is also known to be someone who brings about their own death, through a flaw within their character or through their actions. Proctor's downfall in the play is initiated by a human error, which to a great extent qualifies him to be the tragic hero. Is John Proctor a tragic hero? In this essay I will be exploring his character, personality, and relationship towards others. The Crucible, written by Arthur Miller, depicts the story of the Salem witch-hunt and the chaos it caused. One of the main characters is John Proctor. Proctor is put through many life-changing decisions. In many cases, a decision he made in one situation led to another problem. Miller sets ‘The Crucible’ in a Puritan community in Salem, Massachusetts, in the spring of 1692. The Puritans were a group of religious separatists who sought to worship God in uncomplicated and basic ways. They built their own houses, made their own furniture, and did... ...himself harshly. Before Abigail came and ruined his peace, he was always sure of himself. After Abigail, he was sure that nothing he would ever do would be pure and honest again. Proctor’s â€Å"tragic hero† status was illustrated by his efforts to save his wife from being put to death, his attempt to prove the children are making fraudulent claims, and his unwillingness to confess to practicing witchcraft when accused. Personally I believe all humans are created with flaws, which control our emotions and often twist our lives. We are put into predications because we are having a hard time of controlling them; therefore we just let them flow. John Proctor’s flaws were his faith, the belief of his own dignity, and his pride, that eventually put in to death. He is a tragic hero that dies of his own flaw, which brought up our sympathy and our fear of death.

Defining the Life of Dr. Jekyll and Mr. Hyde      The focal point of this essay is to define the life of Dr. Henry Jekyll, and the transformation he went through in becoming Edward Hyde. Dr. Henry Jekyll is a physician in London.   He is very well respected and is currently experimenting the dual nature of mankind. Edward Hyde is a manifestation of Dr. Jekyll's personality. A large crowd had gathered around and they saw the man, Edward Hyde. The crowd forced the man to give money to this girl for trampling over her. Hyde did not run over her for any reason. He just did it out of spite and evil. He represents all the evil in the world. The reaction of others to him is one of horror because while looking at him, others feel a desire to strike out at him and kill him. His physical appearance brings out the worst evil in other people. Since Hyde represents evil, he is symbolically represented as being much smaller than Dr. Jekyll.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   I believe Dr. Jekyll created Hyde because he had a theory that man has a good side and a bad side. While investigating this, he developed a potion that could release the evil in a person in the form of a totally different person. Then this person could commit any evil act it wanted, and then drink the potion to return back to normal. The only problem with this is the fact that he drank this potion so many times that he was no longer able to control this process. He was unable to transform back into Dr. Jekyll.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   Another example of Hyde's evil is in the killing of Sir Danvers Carew. Sir Danvers appears to have been killed for no apparent reason. The murder of Sir Danvers was seen by a maid who was working nearby. She states that Hyde meet with a man in the street. After the two exchanged words, Mr. Hyde lifted his heavy walking stick and clubbed the old man to death.

1.Intentional torts share three common elements as described in the text on page 113: There must be a volitional or willful act by the defendant.The person so acting must intend to bring about the consequences or appear to have intended to bring about the consequences.There must be causation. The act must be a substantial factor in bringing about the injury or consequences. Examples of intentional tort include: assault, battery, false imprisonment, conversion of property, trespass to land, and intentional infliction of emotional distress. Threatening a patient with an injection or with starting an IV line is an example of assault. Quasi intentional torts differ from intentional torts because with quasi intentional torts the intent is lacking but there is still a volitional act and direct causation. Examples of quasi intentional torts include invasion of privacy and defamation. An example of defamation is when a nurse makes false chart entries about a patient’s lifestyle or diagnoses. 2. All five RN’s that I interviewed regarding their understanding of torts were mostly confused. All five of them were asked if they could tell me the difference between intentional and quasi intentional torts. The first nurse stated that she had no clue as to what I was talking about. The second nurse said, â€Å"I think it has something to do with how nurses treat their patients.† The third nurse just looked at me in a very confused manor, as if she were waiting for me to give her more information. The fourth nurse said, â€Å"I kind of remember learning about that in nursing school, but that was many, many years ago, and I don’t recall exactly what it is. The final nurse knew a little bit more than the others and stated, â€Å"Aren’t they laws that nurses should abide by?† Although, her response was in the form of a question, at least she knew it had something to do with the law. 3. After interviewing these five nurses if became clear to me that nurses need to be reminded or re-educated often about the laws that could affect their practice. An employer providing mandatory continuing education units on torts is one good way of increasing nurses understanding of torts. Another idea could be to place posters in the elevators that nurses might use frequently. These posters should be eye-catching with graphs, diagrams and illustrations showing the differences in torts. Implementing a short video during the orientation process could be another way to increase nurses understanding of torts. Finally, with all the mobile devices that are available these days, I think it would be great if someone created an app that could be downloaded by nurses to their own personal smart phones or tablets explaining the differences between intentional and quasi intentional torts.

Word Choice Allusion vs. Illusion Word Choice: Allusion vs. Illusion â€Å"Allusion† and â€Å"illusion† are both fairly rare words. They also sound similar, making it is easy to confuse them if you’ve never seen them written down. But these terms have very different meanings, so make sure your work is error free by checking out our guide to how they should be used. Allusion (Indirect Reference) An â€Å"allusion† is an indirect reference to something. For example, the title of David Foster Wallace’s novel Infinite Jest is taken from a line in Hamlet. This is an â€Å"allusion† to Shakespeare – rather than a direct reference – because Wallace does not mention Shakespeare or Hamlet in the novel. And while â€Å"allusion† is a noun, the verb form of this word is â€Å"allude†: The title of the novel alludes to Act 5, Scene 1 of Hamlet. Again, the key here is the idea of referencing something indirectly. Illusion (Mistaken Perception or Belief) The word â€Å"illusion† usually refers to a mistaken sense perception, such as a mirage. As such, we could say something like: He saw the hazy outline of an oasis, but he knew it was an illusion. This idea of seeing or sensing something that isn’t really there is also why we say magic tricks are â€Å"illusions,† as well as why we call trick images optical illusions. But we can also use â€Å"illusion† to describe a false belief: I was laboring under an illusion to think I could run a marathon. Here, â€Å"laboring under an illusion† means acting on a mistaken belief. Summary: Allusion or Illusion These words have many letters in common, but they differ in meaning: An allusion is an indirect reference to something. An illusion is a mistaken sense perception or belief. One trick to remembering this difference is the phrase â€Å"Optical illusions will make you ill.† This can remind you that â€Å"illusion† starts will an â€Å"i,† like the word â€Å"ill.† But if you’d like to make extra sure your writing is always error free, you can always ask a proofreader.

Battle of Kwajalein in World War II The Battle of Kwajalein occurred January 31 to February 3, 1944 in the Pacific Theater of World War II (1939-1945). Moving forward from victories in the Solomons and Gilbert Islands in 1943, Allied forces sought to penetrate the next ring of Japanese defenses in the central Pacific. Attacking into the Marshall Islands, the Allies occupied Majuro and then commenced operations against Kwajalein. Striking at both ends of the atoll, they succeeded in eliminating the Japanese opposition after brief but fierce battles. The triumph opened the way for the subsequent capture of Eniwetok and a campaign against the Marianas.   Background In the wake of the American victories at Tarawa and Makin in November 1943, Allied forces continued their island-hopping campaign by moving against Japanese positions in Marshall Islands. Part of the Eastern Mandates, the Marshalls were originally a German possession and were awarded to Japan after World War I. Considered part of the outer ring of Japanese territory, planners in Tokyo decided after the loss of the Solomons and New Guinea that the islands were expendable. With this in mind, what troops were available were shifted to the area to make the islands capture as costly as possible. Japanese Preparations Led by Rear Admiral Monzo Akiyama, Japanese forces in the Marshalls consisted of the 6th Base Force which initially numbered approximately 8,100 men and 110 aircraft. While a sizable force, Akiyamas strength was diluted by the need to spread his command over the entirety of the Marshalls. In addition, many of Akiyamas troops were labor/construction details or naval forces with little ground combat training. As a result, Akiyama could only muster around 4,000 effectives. Believing the assault would strike one of the outlying islands first, he positioned the bulk of his men on Jaluit, Mili, Maloelap, and Wotje. In November 1943, American airstrikes began whittling down Akiyamas air power, destroying 71 aircraft. These were partially replaced over the next several weeks by reinforcements flown in from Truk. On the Allied side, Admiral Chester Nimitz originally planned a series of assaults on the outer islands of the Marshalls, but upon learning of Japanese troop dispositions through ULTRA radio intercepts altered his approach. Rather than strike where Akiyamas defenses were strongest, Nimitz directed his forces to move against Kwajalein Atoll in the central Marshalls. Armies Commanders Allies Rear Admiral Richmond K. Turners 5th Amphibious Force to deliver Major General Holland M. Smiths V Amphibious Corps to the atoll where Major General Harry Schmidts 4th Marine Division would assault the linked islands of Roi-Namur while Major General Charles Corletts 7th Infantry Division attacked Kwajalein Island. To prepare for the operation, Allied aircraft repeatedly struck Japanese airbases in the Marshalls through December. This saw B-24 Liberators stage through Baker Island to bomb a variety of strategic targets including the airfield on Mili. Subsequent strikes saw A-24 Banshees and B-25 Mitchells mount several raids across the Marshalls. Moving into position, US carriers began a concerted air offensive against Kwajalein on January 29, 1944. Two days later, US troops captured the small island of Majuro, 220 miles to the southeast, without a fight. This operation was conducted by the V Amphibious Corps Marine Reconnaissance Company and 2nd Battalion, 106th Infantry.    Coming Ashore That same day, members of the 7th Infantry Division landed on small islands, dubbed Carlos, Carter, Cecil, and Carlson, near Kwajalein to establish artillery positions for the assault on the island. The next day, the artillery, with additional fire from US warships, including USS Tennessee (BB-43), opened fire on Kwajalein Island. Pummeling the island, the bombardment allowed the 7th Infantry to land and easily overcome the Japanese resistance. The attack was also aided by the weak nature of the Japanese defenses which could not be built in depth due to the islands narrowness.   Fighting continued for four day with the Japanese mounting nightly counterattacks. On February 3, Kwajalein Island was declared secure. Roi-Namur At the north end of the atoll, elements of the 4th Marines followed a similar strategy and established fire bases on islands dubbed Ivan, Jacob, Albert, Allen, and Abraham. Attacking Roi-Namur on February 1, they succeeded in securing the airfield on Roi that day and eliminated Japanese resistance on Namur the next day. The largest single loss of life in the battle occurred when a Marine threw a satchel charge into a bunker containing torpedo warheads. The resulting blast killed 20 Marines and wounded several others. Aftermath The victory at Kwajalein broke a hole through the Japanese outer defenses and was a key step in the Allies island-hopping campaign. Allied losses in the battle numbered 372 killed and 1,592 wounded. Japanese casualties are estimated at 7,870 killed/wounded and 105 captured. In assessing the outcome at Kwajalein, Allied planners were pleased to find that the tactical changes made after the bloody assault on Tarawa had bore fruit and plans were made to attack Eniwetok Atoll on February 17. For the Japanese, the battle demonstrated that beachline defenses were too vulnerable to attack and that defense in-depth was necessary if they hoped to stop Allied assaults.

As a strength empathy helps me identify the areas in which individual learners need assistance and as such, I can attend to individual needs besides attending to the needs of the larger class. Individualized attention helps students be free and express their fears and weaknesses to me as an educator out of trust. Individualized attention is instrumental in improving the performance of learners. Authenticity is my other professional disposition with which I approach my work as an educator. Authenticity helps me reckon that different learners have different needs and as such, I vary my methods and approaches to teaching. This comes with an understanding that the method or approach that works for an individual learner or group of learners may not work for another. I emphasize originality and uniqueness in the execution of my duties and in the delivery of content to learners. Authenticity helps me accept my weaknesses and shortcomings and allows me to express myself with genuineness and honesty (Reid et. al., 2011).

Staphylococcus Aureus: Structure and Function Staphylococcus was first discovered in 1880 by Alexander Ogston. Currently, more than 30 different species of the genus has been identified. The name Staphylococcus was derived from Greek, with the prefix Staphylo referring to bunches of grapes and the suffix coccus referring to granule (16). As the meanings suggest, bacteria from Staphylococcus are circular-shaped and their arrangement resembles bunches of grapes when observed under a microscope. Typically, a Staphylococcus has a diameter of approximately 1ÃŽÂ ¼m (21). The bacterial genus, Staphylococcus, will be isolated and identified in this project. This genus has been chosen to review because of its abundance on the skin of mammals and the pathogenic nature of one of its member, Staphylococcus aureus. Apart from skin infections, Staphylococcus aureus could mutate to form Methicillin-resistant Staphylococcus aureus (MRSA), which shows resistance to antibiotics. In both cases, these give rise to medical implications. In addition, the distinctive features of Staphylococcus aureus have increased the ease to isolate and identify it from other species in the genus via culturing and biochemical tests. The aim of the project is to isolate Staphylococcus aureus from a bundle of cat hairs and verify its identity via microscopic examination and biochemical tests. No human specimen is used due to the potential pathogenic property of the bacterium. It is intended that a pure culture of pathogenic Staphylococcus aureus is obtained. For the purposes of this project, the importance of Staphylococcus aureus to humans, its classification in terms of morphology and physiological properties, methods of isolation with the use of growth media and the technique of streak plating and identification by biochemical tests would be the four objectives to be addressed. Objective 1: Importance of Staphylococcus aureus to humans The importance of Staphylococcus aureus to humans would be outlined by a review of its cell structure, cell physiology and environmental niches, followed by the medical implications of Staphylococcus as a result of these properties. Cell structure As a member of the Bacteria domain, it is expected that Staphylococcus has bacterial cell structure. In other words, it lacks nucleus and membrane-bound organelles. The structural elements in a cell of Staphylococcus should include a cell membrane, cell wall, ribosome and nucleoid (6). Moreover, being one of the five genera from the family of Staphylococcaceae, Staphylococcus possesses specific cellular properties that are unique to this family. In particular, it is a cocci and gram-positive bacterium and this indicates that its cell wall is essentially composed of a thick layer of peptidoglycan (21). In addition to the above structures, Staphylococcus aureus possesses some special cellular structures that distinguish it from other species in the genus. This includes the possession of surface proteins that help attachment to proteins such as the fibronectin and fibrinogen-binding proteins involved in blood clotting (3). This cellular property may explain the pathogenic nature of Staphylococcus aureus, as infections might be caused by invasion via wounds. On the other hand, it is worthwhile to note that Staphylococcus does not have flagella and spores (16). That is to say, Staphylococcus aureus is non-motile. Cell physiology The cell physiology of Staphylococcus covers temperature, pH and oxygen requirements. Most Staphylococcus can grow at 45Â °C, but it is reasonable to predict that its optimal temperature for metabolism would be close to the body temperature of humans, which is 37Â °C (5). Concerning the optimum pH for metabolism, the enzymes in Staphylococcus work best in slightly alkaline medium, with a pH range of 7.4 to 7.6 (16). As for oxygen requirement, Staphylococcus is facultative anaerobic (21). This implies that Staphylococcus can grow regardless of the presence of oxygen, but the presence of oxygen would be more favorable. In the presence of oxygen, Staphylococcus utilizes glucose to carry out cellular respiration to generate energy for metabolism. Oxygen performs the role of a terminal electron acceptor and it is completely reduced to water (8). When oxygen is lacking or absent, Staphylococcus may undergo fermentation and lactic acid is the usual product (21). In the process, glucose is converted into substrate pyruvate, followed by its binding to the cofactor Nicotinamide Adenine Dinucleotide (NAD+) to produce lactic acid (6). Moving on the ways Staphylococcus metabolize, as light is not readily available on skin surface and mucous membranes, it is proposed that Staphylococcus obtain energy via organic chemical compounds. Hence it is regarded as a chemotroph (21). The facultative anaerobic property of Staphylococcus may lead to a deduction that it utilizes organic carbon as the source of electron when oxygen is present. Though some Staphylococcus may use reduced forms of inorganic nitrates to generate electrons, its preference towards an aerobic atmosphere should define it as an organotroph (21). When comes to carbon source, Staphylococcus is a heterotrophy (12). That is to say, it attains its carbon source by utilization of organic substances such as sucrose for synthesis of metabolites (19). To summarize, Staphylococcus should be one of the members of the microbial group, Chemo-organotrophic heterotrophs. Environmental niches The environmental niches of Staphylococcus can be addressed by its interactions with the environment as to where it is found, the type of relationship it forms with other organisms and its capability of undergoing mutation. Staphylococcus is commonly found on the skin and mucous membranes of animals with stable body temperatures, including humans (15). Typically, the skin temperature of humans is approximately 32Â °C, which is reasonably close to the optimal temperature of 37oC (22). This enhances the growth of this microbe on skin. Moreover, the salty environment along skin surface due to the production of sweat may also account for the abundance of Staphylococcus in humans, since its enzymatic activity is optimal at more alkaline pH (17). Staphylococcus aureus specifically colonizes in nasal cavity, larynx and on the skin surface of humans (2). The colonization of Staphylococcus aureus is principally achieved by fibrinogen-binding proteins adhering to the epithelial cells of the humans and thus this may outline a host-parasitic relationship between Staphylococcus and humans (10). The interactions of Staphylococcus with the environment may also be underlined by mutation, which often occurs with Staphylococcus aureus. An example would be Methicillin-resistant Staphylococcus aureus (MRSA), a Staphylococcus aureus that is resistant particularly to the antibiotic, Methicillin (21). The mutation is caused by an alteration of the methicillin-resistance gene (mec A) coding for a penicillin-binding protein (4). This results in failure of antibiotics to cure infections caused by Staphylococcus aureus, which will be addressed in the medical implication section. Medical implications of Staphylococcus The features as in the cell structure, cell physiology and environmental niches of Staphylococcus can pose a great diversity of medical implications, which presents the importance of this bacterial genus. Statistics show that Staphylococcus aureus is present in 79% of healthy people (14). Though Staphylococcus may colonize on the skin surface of the host without causing any harms, its ubiquity can still present various medical issues. The MRSA mentioned previously would be one of the problems associated with Staphylococcus. Apart from methicillin, MRSA could show resistance against many other antibiotics such as penicillin and amoxicillin (1). The ineffectiveness of existing antibiotics to cure MRSA infections has resulted in fatality, and it is usually characterized by the incidence of septic shock and pneumonia (11). A rapid increase of MRSA infections has been observed over the decades. The rate of hospitalized MRSA infections was only 2% in 1974 but this figure increases dramatically to approximately 40% in 1997 (13). Consequently, this causes deaths of 19000 in the United States of America annually (11). As Staphylococcus colonies on skin surfaces and mucous membrane, skin infections and diseases associated with mucous membranes could be another medical implication. It is known that Staphylococcus aureus may cause Scalded Skin and Toxic Shock syndromes. Moreover, it may cause urinary tract infections and food poisoning (9). Objective 2: Classification of Staphylococcus The classification of Staphylococcus can be reviewed in terms of its morphology and some of the physiological properties stated above. Morphology The morphology of Staphylococcus can be described as cocci gram positive bacteria arranged in a cluster, which can be readily observed under microscope with the application of gram stain. A purple color would be observed. The reason for its cluster formation may be explained by its capability of undergoing binary fission in multiple planes with daughter cells remains proximal to each other (16). Physiological properties In terms of thermal requirement, Staphylococcus is classified as a mesophile. Regarding pH requirements, it falls into the category of neutrophile. Moreover, being a facultative anaerobe, Staphylococcus is catalase positive and it is generally considered a chemoorganotrophic heterotroph. In addition, Staphylococcus aureus is coagulase positive but not for other species in the genus. The absence of flagella indicates that Staphylococcus is a non-motile bacterium. Objective 3: Methods of Isolation of Staphylococcus The methods of isolation of Staphylococcus would include growing in medium followed by streak plating. Growth media To ensure optimum growth of Staphylococcus colonies, the sample of cat hairs should be enriched in nutrient broth with sodium chloride (NaCl) before plating on a nutrient agar. A nutrient broth normally consists of beef extract and peptone as fuels for growth (21). The temperature of incubation should be 37Â °C and the duration of incubation should be at least a day (20). This ensures that the Staphylococcus isolated can have sufficient time to grow at its optimum temperature. The addition of salt allows for a selective medium for Staphylococcus as it predominantly grows in salty environment. It also increases the pH of the medium to provide for a more alkaline environment to facilitate growth. Alternatively, a growth medium can be done via a Mannitol salt agar (MSA), which consists of 7.5% of NaCl and phenol red as a pH indicator. The medium is then incubated at 37Â °C for two days (14). MSA essentially acts as both a selective and differential medium. NaCl selects for saline-favored Staphylococcus and the pH indicator differentiates between Staphylococcus aureus and Staphylococcus epidermidis. Differentiation can be illustrated by the fact that Staphylococcus aureus utilizes mannitol in the agar for metabolism, and the generation of acidic product is indicated by a yellow color. However, this phenomenon does not apply to Staphylococcus epidermidis (21). Streak Plating Following enrichment, Staphylococcus in the medium can be transferred to an agar plate with nutrient broth and salt, by employment of aseptic techniques. At the same time, a transfer to an agar plate with only nutrient broth should be performed as a control set-up. This is to ensure the effectiveness of the selective media because other bacteria could grow on the agar plate if the medium was not set up properly. Afterwards, the plates would be incubated for a week at 37oC for at least a day as in the incubation of sample in the nutrient broth. Plating and incubation should be repeated a few times to make sure that the colonies grown are pure. Objective 4: Identification by biochemical tests The identity of Staphylococcus cannot be confirmed by carrying out the gram reaction alone due to the fact that a great variety of bacteria from other genus may also show gram positive reaction. Therefore, some biochemical tests have to be performed to verify that the bacteria isolated is in the genus of Staphylococcus and it is of the species Staphylococcus aureus. The catalase, Hugh and Leifsons oxidation fermentation and cogulase tests are regarded as the standard tests for identification of Staphylococcus aureus (18). The mechanism of the tests is outlined below. First of all, as Staphylococcus aureus is facultative anaerobic, it is expected that it contains enzymes to break down harmful products generated along the pathways of aerobic respiration. For instances, catalase breaks down superoxide radical hydrogen peroxide (H2O2) to oxygen and water, which are less harmful (8). Therefore colorless gas bubbles can be observed when H2O2 is added to a colony of Staphylococcus aureus. Moreover, this property allows the Hugh and Leifsons oxidation fermentation test to be performed. The bacterial sample is inoculated in a tube of Hugh Leifsons medium for five days to generate an anaerobic environment (18). As Staphylococcus can undergo fermentation in the absence of oxygen, growth can be observed throughout the tube. At the same time, it is necessary to implement positive and negative controls in order to confirm results. This can be achieved by inoculating bacteria that are known to be fermentative and oxidative respectively in the Hugh Leifsons medium along with the sample of Staphylococcus aureus. Furthermore, the identification test between Staphylococcus aureus and other bacteria in the genus would be based on its reaction with coagulase. Staphylococcus aureus readily coagulates plasma but not for others species in the genus (21). To ensure accuracy of the test, it is preferable to test on colonies extracted from culture plates that are known to contain coagulase positive Staphylococcus aureus and coagulase negative Staphylococcus epididymis respectively. The former acts as a positive control, while the latter acts as a negative control. Conclusion In conclusion, Staphylococcus is a bacterial genus that can pose various medical implications and it can be grown, isolated and identified based on its, environmental niches, morphology, physiological and structural characteristics. The aims of isolating Staphylococcus aureus as a pure culture and identifying by morphology and biochemical tests can be addressed by a review of the four objectives as summarized below. Firstly, it is often found on epidermis of animal skins including humans and its ability to metabolize optimally at 37Â °C and at pH of 7.4-7.6 or salty environment makes it a potential pathogen to humans. In particular, the species Staphylococcus aureus can cause a great diversity of diseases and the mutated Methicillin-resistant Staphylococcus aureus could be fatal owing to its resistance to most antibiotics. This increases the efficiency of isolation of Staphylococcus aureus. Lastly, Staphylococcus aureus can be identified by the catalase, Hugh Leifsons oxidation fermentation and coagulase tests. It is expected that bubbling is observed as a positive result in the catalase test. As for the Hugh Leifsons oxidation fermentation test, growth can be observed throughout the tube. In the coagulase test, clumping of plasma is seen as a positive result and this differentiates Staphylococcus aureus from other species in the genus. This proves to be true upon analyzing her life and accomplishments in the nineteenth century. Hannah More was born in 1745 in Stapleton, Bristol, England (Hopkins xiii). Mary Gladys Jones comments on More's life and gives the description of this literary figure as "a Tory who championed the radical causes of anti-slavery and the education of the poor, a Churchwoman who attended a Dissenting Meeting House [only once] and employed Methodist teachers" (Ford ix-x). The two main political issues that More would write about during the course of her life were anti-slavery issues and the promotion of the education of the poor and women. More herself was educated along with her sisters by her father (Damrosch 100). More and her sisters would later found a successful school which threatened many of her contemporaries. "Local authorities felt threatened b y the sisters' assumption of power and were assuaged only by the Mores' manipulation of traditional feminine roles" (Ford xiii). In the curriculum in More's school, she made a conscious effort to "maintain sexual difference and inequality while upgrading or 'masculinizing' the education of women" (xiii).    As a woman, Hannah More was an unusual figure in nineteenth-century England. She was unmarried and very publicly voiced her opinion. After a failed engagement, More sought financial independence (Damrosch 100). More began publishing her works to the general population. Her works were sold cheap, and preachers, missionaries, or landlords handed them out (Damrosch 189). Since More "celebrated the godly, self-disciplined layperson who looked out for the common good" she sold her works very inexpensively (Ford 1). "At a half-penny to one and one-half penny per installment," More's writings became popular amongst the poor in society (128). More had strong political beliefs that explain much of her writing and methods. She had a "long-held belief that aristocratic virtue, whether civic or personal, dictated general morality which, in turn, ensured the soundness of the commonwealth" (xii). Her moral beliefs are directly effected from her religious beliefs. More fought for anti-s lavery laws in hopes that "the abolition of the African slave trade and the observance of Sabbatarian laws, among other measures, would preempt similar divine retribution against the British ruling classes" (84).

Self-Concept :: Psychology, Gender Difference

Before stepping onto the topic of the way we see ourselves, we first have to try to fully understand the definition of self-concept. The classification of self-concept is defined in many different ways by various researchers and practitioners over years. According to Purkey and William (1988), the popularity and attention of self-concept has been raising since decades of ignorance. Self-concept is mainly regarded as the realisation of our own existence; who we are, what is our purpose and how we fit into the society. Self-concept can be defined in a very complex manner. For example, it can be explained as a cognitive representation of oneself that gives coherence and meaning to one’s experience, including one’s relations to other people. It organises past experiences and plays an important role in assisting us to recognise and interpret relevant stimuli in the social environment (Hewstone, Stroebe & Jonas, 2008). Nonetheless, self-concept can also be clarified as simple as a statement where Fromm (1956) describe as â€Å"life being aware of itself.† After understanding the notion self-concept, we then can proceed to use different approaches to search a suitable answer to the question of â€Å"Why do we ‘see’ ourselves in the way that we do?†. In an experiment done by Montemayor and Eisen (1977), children were studied and questioned for their existence. When the children in the experiment were asked â€Å"Who am I?†, the most common answer given are descriptive and are usually about their appearance. For example, â€Å"I have brown eyes, I have brown hair and et cetera†. Children tend to mainly focus on the description of physical features of their body, address, procession and play activities; kids’ self-concept is more concrete as well as less abstract. In comparison, teenagers were observed and were asked the same question. As teenagers with wider knowledge of their existence, they will probably come up with more profound words and not only focus on the physical parts of body but somehow personal beliefs, motivation and interpersonal characteristics. For example, â€Å"I am a human being. I am a moody person, etc†. Consequently, adolescence’s answers seem to be more abstract but less c oncrete. By asking this question to both childhood and adolescence, significant increase can be seen in self-conceptions and categorised in followings: occupational rule; existential, individuating; ideological and belief reference; the sense of self- determination; the sense of unity; interpersonal style; and physical style (Montemayor & Eisen, 1977). We seem and tends to be a creatures f crisis.Rather than accepting the possibility that serious problems are Just over the horizon and is happening, we choose to look for any hope of positive information to ease our fears, but the fact is that hunger is a big problem in the world today and It is equally important that people should inform and get their selves involve on the issue about food scarcity so that they will know who are the ones truly suffering and the serious damage it does to those people.And also for them to avoid on wasting food, because all the extra food can't be Just given out to the needy and hungry entries, but if it could the government still don't have enough transportation and the money to send all those extra food to the suffering people. Introducing an East World to a West World Essay

On June 26, 1982, Pearl Sydenstricker was born in Hillsboro West Virginia, to two American Presbyterian missionaries who primarily served in China (â€Å"Buck, Pearl S.†). At the age of two, Buck and her family moved back to China where she spent the next 6 years of her life adapting the Chinese culture while integrating aspects of her American heritage as well. Buck’s childhood in China stimulated the bicultural traits that she infused into her writing by exemplifying both American and Chinese culture and values. At 15, Buck was sent to a boarding school in Shanghai and continued her schooling at the Randolph Macon Woman’s College in Lynchburg Virginia, where she graduated in 1914 and taught psychology. Later on she finished her schooling at Cornell University receiving a Master of Arts Degree. Buck also taught English at several universities throughout her career including the University of Nanking, Chung Yang University, and Southeastern University (â€Å"Works of Pearl Buck†). A notable historic event that impacted Buck’s life and her writing was the Boxer Rebellion (â€Å"Works of Pearl Buck†). The boxers were a group of Chinese people who opposed the spread of Western ideas and technology and convinced the empress to drive out all foreigners in China in order to sustain China’s ancient culture and history. Despite the empress’ support for the arising Western influence, the Boxers lashed out by imprisoning the empress and stimulating the attacks on foreigners. This ultimately caused Buck’s family to flee from China back to America in order to escape the rebellions plotted against foreigners (â€Å"A Life Lived in Interesting Times†). Buck also witnessed the foot-binding and murder of female babies in China due to the patriarchal nature that was indigenous to the Chinese culture. This influenced her to become an advocate for woman’s rights and her experience with the Boxer Rebellion profoundly affected her perspective for equality and the political barriers that seemed to separate the values of the West and Asia which was reflected in the various themes she covered. Throughout her life Buck was an outspoken feminist and equalist. She displayed her advocacy by supporting various organizations who strived for equality, promoting birth control, and even adopting nine children. (â€Å"Pearl S. Buck: A Cultural Biography†). She married John L. Buck, and they had one daughter who was mentally disabled. Her daughter’s disability motivated her to support children’s advocacy which inspired her to write â€Å"The Child Who Never Grew† in 1950. It also led her to establish the Pearl S. Buck foundation which strives to assist in the adoption and well-being of Amerasian children. However, her and her husband divorced in 1935 and she soon married Richard J. Walsh, her publisher who she stayed married to until the end of her life (â€Å"Buck, Pearl S.†). Pearl Buck was known for her works such as Dragon Seed which focuses on a female empress named Tzu Hsi who was faced with ruling China during a period of war. Some of her works also reflect her lifestyle that was influenced by two entirely different cultures. Buck’s memoir My Several Worlds tells about her long journey growing up in an environment of political struggle and internal warfare between her own people when her and her family was outcasted by the Chinese as foreigners. It tells about her career as an influential writer, an ambassador, and civil rights activist with her motives defined by the countless experiences of her life. Her most notable title which earned her a Pulitzer Prize and a Noble Prize in Literature was The Good Earth. The book tells of a poor farmer named Wang Lung and his wife who struggle with the changes that war and a corrupted government have brought upon the nation such as famine and poverty. This theme of personal obstacles can most likely relate to Buck’s audience when the book was published in 1931: the many Americans who struggled with the shifting politics and economy during the Great Depression (â€Å"The Good Earth, 1931,† The American Novel†). Buck’s work conveys the complexity of Chinese culture and life to America based off her experience growing up in a bicultural environment (â€Å"The Good Earth,† Barnes and Noble). Buck’s work can best be categorized by the movement of Realism because of the reality that she tries to depict in order to relate the hardships that both the Chinese and Americans share. Realism mainly focused on portraying average experiences and it transitioned after the movement of Romance. It is thought to be known as â€Å"the faithful representation of reality† (â€Å"Realism in American Literature, 1860-1890†). The realist nature of her books forged a connection through the similarities in lifestyle and adversities that the Americans shared with the people of China (â€Å"1860s-1900s Realism,† PBS). She aimed to depict the daily lives of the civilians of China through her characters’ lifestyles that gave an insight of life shrouded by Chinese culture and beliefs. Her ability to express the personal struggles of both the upper, middle, and lower class allowed her to convey the theme of cumulative struggles and experiences of a country as a whole. Pearl Buck’s main purpose for her writing was to link the alien and foreign culture of China to the people of America who were clueless to the cultural values that China imposed (The American Novel). She delivered with a style that precisely aimed to reveal the plain and blunt truth of life, no matter what social class a person belonged to. The works of Pearl Buck have left a lasting impact on both America and China. She created countless milestones in literature such as being one of the first women ever to receive a Noble Prize in Literature and a Pulitzer Prize, and acting as a mediator and uniting the two cultures of America and China through the various situations that she presents of life in China through her characters and various stories. I think that the reason she has left such a profound mark on literature is because of her ability to establish a connection with the audience. Being bi-cultural, Buck utilizes her first-hand experiences of life in China such as war, famine, and even traumatic murder experiences in order to fully convey the events that were occurring in China with the Great Depression of the United States. What makes her writing and themes so universal is that, it portrays the life experiences of people, with personal struggles and I think her themes have the capability to relate to any time period because everyone is able to relate to the hardships of others. Her ability to successfully connect with her audience on a personal level of experience with a completely foreign culture establishes her as one of the most impactful authors in literature in which modern-day writers aspire to achieve. â€Å"Buck’s Accomplishments have proven unique in military, political and literary history† (Taylor). Some even think that â€Å"We are very much in need of a twenty- first century Pearl Buck† (â€Å"A Life Lived in Interesting Times†). This further demonstrates the lasting influence that Buck’s literary work left during her time period, which some believe that â€Å"no one will ever be able to do again† (Taylor). Unfortunately, Pearl Buck died on March 6, 1973 of lung cancer at her home in Danby, Vermont (â€Å"Pearl S. Buck Biography†). Her work continues to live and is enjoyed by people all over the world today. Her deeds as an activist who strived for fairness among people and cultures are survived by the many people who are influenced by her contributions toward equality. She is recognized as a prominent author because of the major impact that she worked towards her whole life so that America would be able to share a culture and history with China. Despite her death, Buck has left her mark as a humanitarian and advocate who had the best interests of people in mind and a legendary writer who brought to America, a style of writing that will remain timeless in practically every age of literature. This gives a circular quality to the story and reminds the reader that on one layer, there are these people of royalty who are in fact subject to voyeurs. Princess Salome is the object of her stepfather Herod’s voyeurism as well as the voyeurism of the page and the Syrian. Beyond this layer, the voyeurs are subject to the moon itself as it looms overhead as a distant but present voyeur. The moon in fact, is the voyeur watch ing this entire story unfold. Getting back to the nature of the story’s conflicts, it is both an incestual situation materializing which leads to the brutal murder and sadistic handling of a man of God. Sun Yatsen's Revolution of 1911 - Essay Example The post Sino-Japanese War (1894-1896) further made the imperial China relinquish more territories including loss of suzerainty of Korea, Taiwan and Manchuria (Lee and Hock 20). The fulcrum of the revolution was the Wuchang rebellion on October 10, 1911, which mainly pushed for changes through the Qing court. The revolution aimed at transforming changing the authoritarian imperial rule to a constitutional monarchy. Led by Sun Yat-sen, a new leadership came into place with him taking the president’s position. The revolution concluded on February 12, 1912 with the abdication of the â€Å"Last Emperor† Puyi. The three people’s principle was responsible for ensuring equality, national identity and establishment of responsive policies as developed by Sun Yat-sen. China’s nationalism or People’s National Consciousness got ideologically drawn from diverse sources including Chinese American thinking, Marxism, Russian ethnological thought and American progressivism (Lee and Hock 28). Nationalism aimed at transforming China’s prosperity by overthrowing the Manchus and limiting the influence of foreign imperialism. Democracy was to proclaim the people’s rights by establishing a republic governed by a constitutional monarchy and not the imperial system. Undoubtedly, nationalism and people’s rights became his primary principles where people had to own the rights of Referendum, Initiative, Recall, and Election. Subsequently, democracy separated the different roles of the government incorporating five powers of the government of Legislation, Judicial, Execu tive, Examination and Control ((Lee and Hock 30). Socialism or people’s livelihood as a principle came after adoption of democracy and nationalism. Andrew Marvells poem To His Coy Mistress Essay In this essay I will compare and contrast Andrew Marvells poem, To His Coy Mistress, with Elizabeth Barrett Brownings sonnet, How Do I Love Thee? Andrew Marvells poem is about an older man trying persuade a younger women to carpe diem (seize the day), in order to make love to her, by using compliments and flattery, Vaster than empires, and should go to praise.'(Stanza 1, line 12) Additionally, Elizabeth Barrett Brownings sonnet is about a female who is expressing her feelings towards a male. Judging by the poem, the woman is deeply in love with the man in a spiritual sense, I love thee to the depth and breadth and height, my soul can reach. (Line 2-3) To His Coy Mistress is a comparatively long poem of 46 lines, which is divided into three stanzas, representing different parts of the argument for which he is trying to persuade her to sleep with him. In the first stanza, the man flatters the women by using grandiose imagery and hyperbole. He says that her coyness would be of no consequence had we but world enough and time (Line 1) and then follows with more detail in the following stanzas. The older man also shows how interested he is by expressing the magnitude of his feelings, by explaining how he would, love you ten years before the flood (Line 8), even if his love were to remain unrequited, till the conversation of the Jews.'(Line 10) In addition he then describes how long he would be prepared to appreciate all of her physical attributes, two hundred to adore each breast, But thirty thousand to the rest.'(Line 15-16) On one hand, he is trying to prove to the young women how much he wants her; on the other hand she could perceive his words in the wrong way, maybe he just wants to sleep with her? You could argue that this is satirising the kings court, because even though he is flattering her, it is inappropriate to assume that he would be allowed to look at her for this long period of time. In the second stanza, he continues to us the grandiose imagery from the first stanza, but introduces a sense of urgency. When he says, but in the very first line, the audience realises that there is going to be a shift in focus. He now says that he hears times winged chariot hurrying near.'(Line 22) From this point, his imagery becomes increasingly desperate. He tries to shock the women into sleeping with him, by talking about her coyness, and if she perseveres in life that way, she runs the risk of only wormstrying that long preserved virginity.'(Line 27-28) He says that his attitude will turn to dust and into ashes all my lust.'(Line 30) This sentence makes the audience more aware of his actual intentions by saying the word lust rather than love, making us believe that he has betrayed his true motives, and slipped up in front of the woman, he is making out he loves. In the last stanza of To His Coy Mistress, it sees him almost demand that they make sport (love). The imagery on this stanza becomes more erotic, and may have more than one interpretation. Let us roll.up into one ball, and tear our pleasuresthrough the iron gates of life.'(Line 41 and 44) Rolling up into a ball could represent two people making love together or a cannon ball which will smash down, the iron gates of life. These gates could represent the womans chastity belt or societys conventions which would frown upon an extramarital affair. The last two lines are similarly ambiguous. They both tie in with lines from the second stanza. Thus, through we cannot make our sun, stand still, yet we will make him run (Line 45 46), suggests that they should make the most of their time they are spending together, and conceive a child (son). Elizabeth Barrett Brownings poem, How do I love thee presents quite a contrast to Andrew Marvells poem. Its imagery is humble, and very personal. It is an expression of deep love and devotion from one person to another. The purpose of the poem is to quantify the dimensions of her love and at the beginning of the poem, it is very clear to the audience that this woman is deeply in love with her partner By starting with the line, How do I love thee? Let me count the ways'(Line 1) she begins to compare her love to religion and emotions rather than physical attributes, I love thee freely, as men strive for right. (Line 7) The imagery she uses to supposedly quantify this love is suggestive of infinity, and a love which has no boundaries, even after the soul is free from the physical body, I shall but love thee after death. (Line 14) Constructive descriptions are always used in this poem, compared to Andrew Marvells, which also describes the negative issues. Besides Brownings poem having a very romantic meaning, you could question why the woman needs to prove her love for her partner, because she continually mentions and repeats, I love thee in most of her sentences. She could be indeed counting the ways she loves her partner, however maybe her partner needs reassuring of her love for him. One could argue that, Barrett Browning chose the sonnet form for this poem, for the purposes of contrast; to take something which is supposedly infinitive (her love) and place it in a finite and restricted form (sonnet). Alternatively, people could think different because there are such strict rules governing sonnet writing, (i.e. 10 syllables per line, 14 lines etc) and people might question why she chose such a rigid format, for something which she feels most strongly about. Overall, the two poems are opposites, but they are both concerned with the concept of time, human life and love. At the end of the poem, browning says, if god choose, I shall love thee better after death,'(Line 13-14) showing that even after she dies, her love for this man will continue to grow, loving him for eternity. To His Coy Mistress, and How Do I Love Thee have many correlations with each other. For example; Both poems are about love; but represent different kinds of love. Andrew Marvells poem is about lust and sexual gratification, while Elizabeth Barrett Brownings poem is about true love and loyalty. The sonnet is written from a womens point of view, where she expresses her true love for her husband. The other poem is written from a mature mans point of view, and represemts his lust for a younger women. This poem is an elaborate chat up line to present a logical argument in order to persuade her to make love to him. This can show how the significance of a poem can differ because of the century it has been written in. To his coy mistress, was written in the 1600s while How do I love thee was written in the 1800s. The cultural difference between these two periods, is the writing style, as in the 1600s people were interested in composing clever arguments, and were more interested in writing about sex, lust and passion. Whereas in the 1800s, the poems written had more true meaning, with a deeper, more romantic feeling. And into ashes all my lust, compared to, How do I love theeLet me count the ways. Many love poems are written in a very traditional format, with very strict rules. Elizabeth Barrett Browning took the challenge to write about something which is supposed to have no limits (love) into something which is restricted (sonnet). On the other hand Andrew Marvells poem is all based around carpe diem, in a non-traditional format, with no strict rules or guide-lines. This can affect the imagery used as one has no limitation to the amount of syllables, lines or stanzas, while the other has a restricted format leaving a certain amount of phrases which can be used. Clearly this means that there are different types of imagery used between the two poems since, How do I love thee uses abstract and emotional imagery, which tends to be highly personal and humble. In some cases you cannot quite put a finger on what she is describing. I love thee to the level of every day, most quite nearby sun and candlelight. (Line 5-6) To His Coy Mistress, tends to use ostentatious hyperbole and grandiose imagery. He uses big overdone, tacky images, by using phrases such as, times winged chariot, instant fires and iron gates of life. You can picture what the man is describing; but on the other hand, you can tell that he might be misleading the audience to make them believe that he is in love with this woman. Throughout the whole of my essay, I have come to the conclusion, that both poems have many comparisons, similarities and differences. They both describe a form of love between two people. In my opinion, the poem I prefer is, To his coy mistress, because it had more of an effect on me, because of the language used. The man comes across as very desperate, but on the other hand with very good charisma. The imagery used is effective, as I could relate and imagine what he is describing. As well as keeping the audience intrigued on what the women will do next, he leaves the ending on a cliff hanger, by not telling on the final decision the women makes! However, I do not dislike the poem, How do I love thee because it has appropriate language for the concept she is describing. I just think it is rather tedious describing only the optimistic qualities about a person, for the reason that everybody has faults and pessimistic attributes. Furthermore, I didnt find it unique or distinctive from other love poems, since the language and imagery was very similar.
